WASHINGTON (Reuters) - Americans on Tuesday marked the 40th anniversary of the Supreme Court decision that legalized abortion, even as battles over the contentious issue have largely shifted from the federal courts to statehouses.
Dozens of protesters braved frigid temperatures to gather in front of the Supreme Court on the anniversary of the landmark January 22, 1973, Roe v. Wade ruling that made abortion legal in the first three months of pregnancy. With a companion ruling, the decision declared abortion a constitutional right.
Anti-abortion activists spread 3,300 flowers out on the Washington sidewalk to represent the number of abortions they said took place daily in the United States.
Away from Washington, about two dozen abortion rights activists rallied in front of Mississippi's sole abortion clinic, the Jackson Women's Health Organization in Jackson.
The supporters, including the National Organization of Women, said they wanted to celebrate the ruling and show that the fight to preserve women's rights continued.
"Choice is good," said Alex McInick, 18, a student at Millsaps College in Jackson. "Nobody should be able to tell someone what they should do to their body."
Across the street, Roy McMillen, 69, an anti-abortion activist, said the procedure led to social ills that cost everyone.
Surrounded by graphic posters of aborted fetuses, McMillen said, "The worst thing that happened in the 20th century was the advent of birth control and the legalization of abortion."
The protests and others this week come in the wake of a Pew Research Center poll which found that most Americans remained opposed to overturning the decision, with opinions little changed over two decades.
"Millions of women have been bruised and diminished. Our country has violated the principles on which it was founded," the Reverend Patrick Mahoney of the Christian Defense Coalition told reporters outside the Supreme Court.
Mahoney was nearly drowned out by a handful of abortion rights activists who chanted "Abortion without demand and without apology" and profanity-laced slogans.
Mahoney said the anti-abortion movement had been energized by the re-election of President Barack Obama, who favors abortion rights and backs Roe v. Wade.
Despite his abortion rights stance, Obama has been a "silver lining" since he had helped move anti-abortion campaigns to the state and local level, away from federal policy-makers, he said.
The highlight of Washington events is expected to be a March for Life rally near Capitol Hill on Friday, with former Republican presidential candidate Rick Santorum among the scheduled speakers.
The rally, which has drawn thousands of protesters in past years, will be followed by a march on the Supreme Court building.
Separate prayer services also are scheduled in Washington by the National Pro-Life Religious Council and the U.S. Catholic bishops' Committee on Pro-Life Activities.
Among other anti-abortion events, the conservative Family Research Council is hosting on Friday its yearly ProLifeCon, which gathers bloggers, activists and lawmakers.
Abortion rights campaigners have few Washington events scheduled around the anniversary, with NARAL Pro-Choice America promoting "Blog for Choice Day" on Tuesday.
The Guttmacher Institute, a reproductive health and rights organization, said this month that 2012 brought the second-highest number of state-level restrictions, trailing only 2011.
"More than half of all U.S. women of reproductive age (15-44) now live in a state that is hostile to abortion rights, whereas fewer than one-third did a decade ago," the group said in a statement.
Recent Washington fights over reproductive rights have centered on Obama's healthcare law. The Senate in March rejected a Republican measure that would have let employers opt out of birth control coverage and other services on moral grounds.

We wax, we wane. It's the dance of life.

Everything alive will eventually die, we know that, but now we can read the pattern and see death coming. We have recently learned its logic, which "You can put into mathematics," says physicist Geoffrey West. It shows up with "extraordinary regularity," not just in plants, but in all animals, from slugs to giraffes. Death, it seems, is intimately related to size.
Life is short for small creatures, longer in big ones. So algae die sooner than oak trees; elephants live longer than mayflies, but you know that. Here's the surprise: There is a mathematical formula which says if you tell me how big something is, I can tell you ? with some variation, but not a lot ? how long it will live. This doesn't apply to individuals, only to groups, to species. The formula is a simple quarter-power exercise: You take the mass of a plant or an animal, and its metabolic rate is equal to its mass taken to the three-fourths power. I'll explain how this works down below, but the point is, this rule seems to govern all life.
A 2007 paper checked 700 different kinds of plants, and almost every time they applied the formula, it correctly predicted lifespan. "This is universal. It cuts across the design of organisms," West says. "It applies to me, all mammals, and the trees sitting out there, even though we're completely different designs."
It's hard to believe that creatures as different as jellyfish and cheetahs, daisies and bats, are governed by the same mathematical logic, but size seems to predict lifespan. The formula seems to be nature's way to preserve larger creatures who need time to grow and prosper, and it not only operates in all living things, but even in the cells of living things. It tells animals for example, that there's a universal limit to life, that though they come in different sizes, they have roughly a billion and a half heart beats; elephant hearts beat slowly, hummingbird hearts beat fast, but when your count is up, you are over. Plants pulse as well, moving nourishment through their veins. They obey the same commands of scale, and when the formula says "you're done," amazingly, the buttercup and the redwood tree obey. Why a specific mathematical formula should govern all of us, I don't completely understand, but when the math says, "it's time," off we go ...
Of course these rules do not tell any particular bee or dog or person when they are going to die. Every individual is subject to accident, caprice, luck. No, this is a general rule. It governs species. Modern humans have managed, because of medicines and hygiene, to become an exception, but 50,000 years ago, we were probably part of the pattern. If you're interested in quarter power scaling, you can check out "Of Mice and Elephants: A Matter of Scale," by George Johnson or go back to an earlier blog post I wrote here. But to summarize, nature goes easy on larger creatures so they don't wear out too quickly. An elephant has trillions more cells than a shrew, and all those cells have to connect and communicate to keep the animal going. In any big creature, animal or plant, there are so many more pathways, moving parts, so much more work to do, the big guys could wear out very quickly. So Geoffrey West and his colleagues found that nature gives larger creatures a gift: more efficient cells. Literally.
The cells in an elephant do more work in a minute than the cells of a mouse. That's why an elephant cell can beat at a slower rate than the rattatat-tat of a mouse cell. Both wear out after a billion and a half beats, but the elephant does it more slowly. As for the peculiar quarter power scaling differences, that rule emerges from the data when you plot the different lifespans of animals or plants on a graph. On the eve of President Barack Obama?s inauguration has been a quiet move to extend his campaign apparatus in the form of a social media Crossroads GPS-like group.
obama1231The Los Angeles Times first picked up on the story on Thursday, followed by Politico. President Obama confirmed the move on Friday, which comes in the form of setting up a 501(c)(4) nonprofit called Organizing for Action.
The Supreme Court?s Citizens United decision in 2010 allows corporations to donate unlimited sums of money to such groups, without their names being made public. On Friday, a White House source told BuzzFeed that Organizing for Action will voluntarily disclose the names of its donors.
Ironically, such 501(c)(4) groups were criticized by the Obama campaign during the 2012 election season, even though there was one group associated with the Obama camp.
The efforts of bigger 501(c)(4) groups like Karl Rove?s Crossroads GPS were more effective at raising large amounts of money?and received heavy criticism from Democrats.
But in the end, President Obama?s social media/marketing machine, based in Chicago, was more effective at spending money and getting out the vote against GOP contender Mitt Romney.
So now, the remnants of that machine will become Organizing for Action and its leadership group will reportedly include such Obama heavy-hitters as Jim Messina (the Obama re-election campaign?s manager), Stephanie Cutter, Robert Gibbs, and David Plouffe.
Vice President Joe Biden dropped hints last week the group will tackle gun control as its first issue.
And when it comes to Organizing for Action, its advocacy of issues will be key to maintaining its nonprofit status.
The primary purpose of a 501(c)(4) group is to engage in the ?promotion of social welfare.? That would include TV ads and social media campaigns about gun control, immigration, or other issues, but not ones targeting politicians or elections.
The PBS series Frontline has a good explanation about how a 501(c)(4) group can be used by smart political operators. It explains that much like George Carlin?s ?Seven Words You Can?t Say on Television,? there are eight words or phrases that a 501(c)(4) can?t use in an ad.
By avoiding the words which are directly related to election campaigns, a group like Organizing for Action can run as many TV and social media campaigns as it can afford (or sees fit to run without annoying too many people).
In an email on Friday to supporters obtained by Politico, President Obama laid out the issues that Organizing for Action will address.
?We?ve got to keep working on growing the economy from the middle out, along with making meaningful progress on the issues we care about ? immigration reform, climate change, balanced deficit reduction, reducing gun violence, and the implementation of the Affordable Care Act,? President Obama said.
There were already reports about two weeks after President Obama?s re-election that an Internet marketing system called Narwhal, used during the campaign, would be used to shape the public debate over the upcoming fiscal cliff negotiations.
Messina spoke at a conference hosted by Politico and described Obama for America, the operation that included Narwhal, a Dashboard program, a team of programmers, and 32,000 volunteers.
?It would be very easy for supporters today to go and start asking people to call members of Congress. ? I am sure you will see our supporters start doing that,? Messina said in November.

Sadly, however, this financial stress has also contributed to the breakdown of marriages and has left many couples facing divorce. While it may seem the two legal issues are independent of each other, the implications of each can have serious legal consequences on the other.
If the debt a couple has incurred is substantial, it may make sense to jointly enter into and complete a bankruptcy before finalizing the divorce. The advantage of discharging debts before the divorce is final can simplify the division and distribution of the marital property and debts. It can also avoid possible future liability of debts assigned to your spouse in the event of his or her bankruptcy.? Sometimes, however, one spouse will file for bankruptcy after the divorce proceedings are under way. This can have serious implications on the divorce process.
Once a petition for bankruptcy is filed, an automatic stay becomes effective and a bankruptcy estate is also created. The estate includes all property of the person filing for bankruptcy?the ?Debtor.? With a bankruptcy pending the court can still move forward to determine custody, child support, and spousal support but any property division decision must wait until the bankruptcy proceedings have concluded or the automatic stay has been properly lifted as to marital assets.
While there are several different types of bankruptcy, usually the only two that come into play in a divorce situation is Chapter 7 bankruptcy and Chapter 13 bankruptcy.
Under Chapter 7 bankruptcy a total discharge of debts is obtained. Under Chapter 13 bankruptcy, a repayment plan is structured by the court allowing the debtor to make payments over time in order for the debtor to be able to keep certain assets and then have remaining debts discharged. The type of bankruptcy affects which debts can be discharged, if and when debts will be discharged, and the contents of the bankruptcy estate. Property settlements between spouses in a divorce action are not dischargeable under Chapter 7 but can be dischargeable if certain conditions are met under Chapter 13. This means that a spouse or ex-spouse filing for bankruptcy could be released from their obligation to pay debt distributed to them by the court in the divorce proceeding. Further, the spouse or ex-spouse can potentially avoid having to make payments to the other spouse if those payments were ordered as part of the property settlement. The spouse not seeking the bankruptcy may become liable for debts that were distributed to the other spouse in the divorce.
Bankruptcy, however, does not discharge a debtor from their child support or alimony obligation. On the contrary, these obligations get priority treatment under the bankruptcy law. This means that under Chapter 7 the first debts to get paid are the child support and alimony obligations of the debtor. Under Chapter 13, any support obligations must be paid in full during the bankruptcy repayment plan. The failure to pay current support obligations in full can result in denial of a discharge under Chapter 13.
Bankruptcy and divorce have serious legal consequences. The issues mentioned in this article highlight the major issues that can arise when bankruptcy and divorce intersect. There are other issues that can arise. You should seek the advice of a competent divorce attorney and a competent bankruptcy attorney when facing these issues.

BALTIMORE (AP) ? Earl Weaver, the fiery Hall of Fame manager who won 1,480 games with the Baltimore Orioles seemingly was engaged in nearly as many arguments with umpires, has died. He was 82.
Dick Gordon, Weaver's marketing agent, said Saturday that Weaver died while on a Caribbean cruise sponsored by the Orioles. Gordon said Weaver's wife told him that Weaver went back to his cabin after dinner and began choking between 10:30 and 11 Friday night. Gordon said a cause of death has not been determined.
The Duke of Earl, as he was affectionately known in Baltimore, took the Orioles into the World Series four times over 17 seasons but won only one title, in 1970. His .583 winning percentage ranks fifth among managers who served 10 or more seasons in the 20th century.
"Earl Weaver stands alone as the greatest manager in the history of the Orioles organization and one of the greatest in the history of baseball," Orioles owner Peter Angelos said. "This is a sad day for everyone who knew him and for all Orioles fans. Earl made his passion for the Orioles known both on and off the field. On behalf of the Orioles, I extend my condolences to his wife, Marianna, and to his family."
Weaver was a salty-tongued manager who preferred to wait for a three-run homer rather than manufacture a run with a stolen base or a bunt. While some baseball purists argued that strategy, no one could dispute the results.
"He was an intense competitor and smart as a whip when it comes to figuring out ways to beat you," said Davey Johnson, who played under Weaver in the minor leagues and with the Orioles from 1965 to 1972.
Weaver had a reputation as a winner, but umpires knew him as a hothead. Weaver would often turn his hat backward and yell directly into an umpire's face to argue a call or a rule, and after the inevitable ejection he would more often than not kick dirt on home plate or on the umpire's shoes.
He was ejected 91 times, including once in both games of a doubleheader.
Asked once if his reputation might have harmed his chances to gain entry into the Hall of Fame, Weaver admitted, "It probably hurt me."
Those 91 ejections were overshadowed by his five 100-win seasons, six AL East titles and four pennants. Weaver was inducted into the Hall in 1996, 10 years after he managed his final game with Baltimore at the end of an ill-advised comeback.
In 1985, the Orioles' owner at the time, Edward B. Williams, coaxed Weaver away from golf to take over a struggling squad. Weaver donned his uniform No. 4, which had already been retired by the team, and tried to breathe some life into the listless Orioles.
Baltimore went 53-52 over the last half of the 1985 season, but finished seventh in 1986 with a 73-89 record. It was Weaver's only losing season as a major-league manager, and he retired for good after that.
"If I hadn't come back," Weaver said after his final game, "I would be home thinking what it would have been like to manage again. I found out it's work."
Weaver finished with a 1,480-1,060 record. He won Manager of the Year three times.
"I had a successful career, not necessarily a Hall of Fame career, but a successful one," he said.
Weaver came to the Orioles as a first base coach in 1968, took over as manager on July 11 and went on to become the winningest manager in the history of the franchise.
"Earl was such a big part of Orioles baseball and personally he was a very important part of my life and career and a great friend to our family," Hall of Fame shortstop Cal Ripken said. "His passion for the game and the fire with which he managed will always be remembered by baseball fans everywhere and certainly by all of us who had the great opportunity to play for him. Earl will be missed but he can't and won't be forgotten."
He knew almost everything about the game. He was also a great judge of human character, and that's one of the main reasons why he was loved by a vast majority of his players even though he often rode them mercilessly from spring training into October.
"His bark was worse than his bite, but you had to know him and kind of grow up with him, and then you loved him like a father," Johnson said. "He was a used-car salesman in the minor leagues during the offseason, so he learned a lot of ways to sell you on just about anything."
Pat Dobson, who pitched two seasons under Weaver, said, "Certainly, the years I played for him were the two most enjoyable years I've had."
During games Weaver smoked cigarettes in the tunnel leading to the dugout and he never kicked the habit. He suffered a mild heart attack in August 1998, and the Orioles' manager at the time, Ray Miller, wondered aloud how his mentor was holding up.
"I wouldn't want to talk to him if he hasn't had a cigarette in 10 days," Miller joked. "They've probably got him tied to a chair."

FOLLOWING a Freedom of Information, it is being reported that the government is clamping down on tax avoidance arrangements through the use of Managed Service Companies (MSCs) legislation.
Says the report, legislation enables the government to recover tax and National Insurance contributions, and has caused investigations to treble in a short period of time. The news will no doubt be of interest to those working within both employment and financial industries, including UK umbrella companies such as Tarpon UK Ltd.
The company which submitted the Freedom of information requests to HM Revenue and Customs (HMRC)? was recruitment law specialist, Lawspeed. It says that the number of MSC tax avoidance arrangement enquiries had risen from 216 to 860, in the space of seven months.
Lawspeed make it clear that since there has been 142 debt transfer notices issued since April 2011, recruitment supply companies that work with service providers should be very cautious.
The MSCs legislation defines Managed Service Companies as any contractor company that pays its worker more than employed levels of income. If HMRC discovers recruiters using companies that chose to avoid tax and National Insurance, this may result in the outstanding tax and National Insurance debt being transferred to them.

AIN AMENAS, Algeria (AP) ? Algerian special forces stormed a natural gas complex in the middle of the Sahara desert on Saturday in a "final assault" that ended a four-day-old hostage crisis, according to the state news agency and two foreign governments. At least 19 hostages and 29 Islamist militants have been killed.
The report, quoting a security source, didn't say whether any hostages or militants remained alive, and it didn't give the nationalities of the dead.
It said the army was forced to intervene after a fire broke out in the plant and said the militants killed the hostages. It wasn't immediately possible to verify who killed the captives.
Seven hostages and 11 militants were killed in Saturday's operation, adding to the previous tally of 12 captives and 18 kidnappers.
The Ain Amenas plant is jointly run by BP, Norway's Statoil and Algeria's state-owned oil company. The governments of Norway and Britain said they received confirmation the siege was over.
The entire refinery was mined with explosives and set to blow up, the Algerian state oil company Sonatrach said in a statement, adding that the process of clearing the explosives had begun. The Algerian media reported that the militants had planned to blow up the complex.
The siege transfixed the world after radical Islamists linked to al-Qaida stormed the complex, which contained hundreds of plant workers from all over the world.
Algeria's response to the crisis was typical of the country's history in confronting terrorists ? military action over negotiation ? and caused an international outcry from countries worried about their citizens. Algerian military forces twice assaulted the areas where the hostages were being held with minimal apparent negotiation ? first on Thursday and then on Saturday.
The latest deaths bring the official Algerian tally of dead to 19 hostages and 29 militants, although reports on the number of dead, injured and freed have been contradictory throughout the crisis. Militants originally said they had seized 41 foreign hostages.
The al-Qaida-linked militants attacked the plant Wednesday morning. They crept across the border from Libya, 60 miles (100 kilometers) away, and fell on a pair of buses taking foreign workers to the airport. The buses' military escort drove off the attackers in a blaze of gunfire that sent bullets zinging over the heads of crouching workers. A Briton and an Algerian ? probably a security guard ? were killed.
Frustrated, the militants turned to the vast gas complex, divided between the workers' living quarters and the refinery itself, and seized hostages, the Algerian government said. The gas flowing to the site was cut off.
On Thursday, Algerian helicopters opened fire on a convoy carrying both kidnappers and their hostages, resulting in many deaths, according to witnesses.
In their final communications, the militants said they were holding seven hostages: three Belgian, two Americans, a Japanese and a Briton. They had threatened to kill them if the Algerian army attacked.
Algerian authorities estimated that about 30 militants occupied the Ain Amenas site Wednesday and with 18 already reported dead, it appeared Saturday that the hostage crisis was finally over.
The standoff has put the spotlight on al-Qaida-linked groups that roam remote areas of the Sahara, threatening vital infrastructure and energy interests. The militants initially said their operation was intended to stop a French attack on Islamist militants in neighboring Mali ? though they later said it was two months in the planning, long before the French intervention.
The accounts of hostages who escaped the complex highlight the cavalier attitude toward their lives taken by both kidnappers and the military.
Ruben Andrada, 49, a Filipino civil engineer who works as one of the project management staff for the Japanese company JGC Corp., described how Algerian helicopter gunships had earlier opened fire on vehicles carrying hostages and the gunmen who used them as shields.
On Thursday, about 35 hostages guarded by 15 militants were loaded into seven SUVs in a convoy to move them from the housing complex to the refinery, Andrada said. The militants placed "an explosive cord" around their necks and were told it would detonate if they tried to run away, he said.
"When we left the compound, there was shooting all around," Andrada said, describing the helicopters' attack. "I closed my eyes. We were going around in the desert. To me, I left it all to fate."
Andrada said when the vehicle he was in overturned, some of the passengers were able to escape. He sustained cuts and bruises and was grazed by a bullet on his right elbow. He later saw the blasted remains of other vehicles, and the severed leg of one of the gunmen.
The site of the gas plant spreads out over several hectares (acres) and includes a housing complex and the processing site, about a mile apart, making it especially complicated for the Algerians to secure the site and likely contributed to the lengthy standoff.
"It's a big and complex site. It's a huge place with a lot of people there and a lot of hiding places for hostages and terrorists," said Col. Richard Kemp, a retired commander of British forces who had dealt with hostage rescues in Iraq and Afghanistan. "These are experienced terrorists holding the hostages."
Casualty figures varied widely. The Algerian government now says 19 hostages and 29 militants have died since Thursday. Before the final assault, the militants claimed through the Mauritanian website ANI that the helicopter attack alone killed 35 hostages and 15 militants.
One American, from Texas, is among the dead, and the militants offered to trade two other American hostages for two terrorists behind bars in the U.S., an offer firmly rejected by Washington. At least one Briton, a Frenchman and Algerians have also died in the standoff. Escaped Algerian workers describe seeing people of many nationalities, including Japanese, shot down.
French Defense Minister Jean-Yves Le Drian said Saturday that there are believed to be no more French hostages in the gas complex. He said the Frenchman killed, Yann Desjeux, was a former member of the French Special Forces and part of the security team. The remaining three French nationals who were at the plant are now free, the Foreign Ministry said.
Britain's Foreign Secretary William Hague confirmed that as of Saturday, there were "fewer than 10" British nationals still at risk or unaccounted for and "the majority of" Britons at the plant were now safe, he said.
Statoil CEO Helge Lund said Saturday that there were only six Norwegians unaccounted for, from the 17 at the plant at the time of the attack.
Romanian Prime Minister Victor Ponta said Saturday one Romanian hostage was killed in the course of the siege, while four escaped unharmed.
The attack by the Mali-based Masked Brigade had been in the works for two months, a member of the brigade told the online Mauritanian news outlet. He said militants targeted Algeria because they expected the country to support the international effort to root out extremists in neighboring Mali and it was carried out by a special commando unit, "Those Who Signed in Blood," tasked with attacking nations supporting intervention in Mali.
The kidnappers focused on the foreign workers, largely leaving alone the hundreds of Algerian workers who were briefly held hostage before being released or escaping.
Several of them arrived haggard-looking on a late-night flight into Algiers on Friday and described how the militants stormed the living quarters and immediately separated out the foreigners.
Mohamed, a 37-year-old nurse who like the others wouldn't allow his last names to be used for fear of trouble for himself or his family, said at least five people were shot to death, their bodies still in front of the infirmary when he left Thursday night.
Chabane, who worked in food services, said he bolted out the window and was hiding when he heard the militants speaking among themselves with Libyan, Egyptian and Tunisian accents. At one point, he said, they caught a Briton.
"They threatened him until he called out in English to his friends, telling them, 'Come out, come out. They're not going to kill you. They're looking for the Americans,'" Chabane said.
"A few minutes later, they blew him away."

There are trying to push the envelope on the issue of Medicare age beyond what has even been discussed so far. From the Business Roundtable:
Increase Retirement Age: The Social Security retirement age should be raised from age 67 to age 70. The unique needs of individuals in in physically demanding occupations should be accommodated and the Social Security Disability Insurance Program should be modernized. [...]
Protect Medicare for Those Approaching Retirement: Medicare?s age of eligibility should be moved to age 70. However, this will not affect those age 55 or older today.
Raising the Medicare age is an unique terrible policy idea. It doesn?t save the federal government much money and by forcing older people into the less cost effective private insurance, it increases overall health care spending. It increases spending for almost everyone including seniors, everyone buying individual insurance, and state Medicaid programs.
Basically the only group of people who would really benefit from this change would be individuals in the health care industry who would see their revenues increase significantly as a result. People like the CEO?s of Aetna, Unitedhealth, Pfizer and Merck who all happen to be members of the Business Roundtable.

Jan. 17, 2013 ? In each cell, thousands of regulatory regions control which genes are active at any time. Scientists at the Research Institute of Molecular Pathology (IMP) in Vienna have developed a method that reliably detects these regions and measures their activity.
Information on the new technology was just published online in the journal Science.
Genome sequences store the information about an organism's development in the DNA's four-letter alphabet. Genes carry the instruction for proteins, which are the building blocks of our bodies. However, genes make up only a minority of the entire genome sequence -- roughly two percent in humans. The remainder was once dismissed as "junk," mostly because its function remained elusive. "Dark matter" might be more appropriate, but gradually light is being shed on this part of the genome, too.
Far from being useless, the non-coding part of DNA contains so-called regulatory regions or enhancers that determine when and where each gene is expressed. This regulation ensures that each gene is only active in appropriate cell-types and tissues, e.g. hemoglobin in red blood cell precursors, digestive enzymes in the stomach, or ion channels in neurons. If gene regulation fails, cells express the wrong genes and acquire inappropriate functions such as the ability to divide and proliferate, leading to diseases such as cancer.
Despite the importance of gene regulatory regions, scientists have been limited in their ability to study them on a genome-wide scale. Their identification relied on indirect means, which were error prone and required tedious experiments for validating and quantifying enhancer activities..
Alexander Stark and his team at the IMP in Vienna now closed this gap with the development of a new technology called STARR-seq (self-transcribing active regulatory region sequencing), published online by Science this week. STARR-seq allows the direct identification of DNA sequences that function as enhancers and simultaneously measures their activity quantitatively in entire genomes.
Applying their technology to Drosophila cells, the IMP-scientists surprisingly find that the strongest enhancers reside in both regulatory genes that determine the respective cell-types as well as in broadly active "housekeeping" genes that are required for basic cell survival in most or all cells. In addition, they find several enhancers for each active gene, which might provide redundancy to ensure robustness of gene regulation.
The new method combines advanced sequencing technology and highly specialized know-how in bio-computing. It is a powerful tool which, according to Alexander Stark, will prove immensely valuable in the future. "STARR-seq is like a magic microscope that lets us zoom in on the regulatory regions of DNA. It will be crucial to study gene regulation and how it is encoded in the genome -- both during normal development and when it goes wrong in disease."
